The invention discloses a method for collecting and measuring the concentration of a one-step virus aerosol, belonging to the field of biomedicine. The method comprises the following steps: (1) the sensitive cells matched with the detected virus aerosol enriched culture, enriched a large number of sensitive cells; (2) to be detected the virus aerosol direct to step (1) prepared in culture sensitive period on the body, culture the virus, and sensitive cells form plaque, plaque number concentration to calculate by detection of virus aerosol. The method of the invention will directly collected virus aerosol enriched sensitive cell culture on the body, immediately after sampling, without elution, transfer of intermediate links, determine the calculated aerosol concentration sampling by the number of virus plaque, to overcome the shortcomings of the existing methods of the indirect sampling method in existence, with rapid and sensitive and accurate reliable, strong anti-interference capability, convenient and economical, simple and practical, no environmental pollution.
