目前在做厌氧产气的微生物菌(主要有肉毒杆菌、产气荚膜梭菌、破伤风梭菌、艰难梭菌等)培养的时候,可采用的方法有:厌氧缸法、气袋法、厌氧手套箱等方法,针对现有的常用气袋法,一般使用透明塑料袋,内装气体发生管(有硼氢化钠的碳酸氢钠固体以及5%柠檬酸安瓿)、美兰指示剂管、钯催化剂管、干燥剂。放入已接种好的平板后,尽量挤出袋内空气,然后密封袋口。先折断气体发生管,后折断美兰指示剂管,袋内在半小时内造成无氧环境。但气袋也有一定的空间范围,厌氧剂市价贵,一个平板放置一个气袋就需使用掉一包厌氧剂来确保厌氧培养环境。若是采用填充CO2的方法进行,需要额外设置装配有单独的二氧化碳的容器管道,且无相关研究报道纯CO2条件适用于厌氧菌的生长。另外很多时候需要一次进行几个重复和/或几个梯度的平板或几种不同标本的培养平板同时培养,若是中途取出其中一个培养平板查看培养结果,其他培养平板就会接触到氧气,对其他培养平板的培养环境造成影响。因此,设计一种具有密封性能好、独立的、厌氧培养过程使用厌氧剂的量少,从而提高厌氧培养效果和效率,且降低厌氧培养成本的培养装置尤为重要。
